Mega Drive version
Super Options
On the main menu press 
(10) START .
UFO
Select a difficulty higher than normal, fire once then wait. If successful, a tiny UFO should fly from the right hand side of the screen, and if hit, increases the chances of getting useful items.
This trick only seems to work on Japanese consoles.
Master System version
Sega logo
Although the Master System version of Dynamite Duke was not released in Japan, there is programming to display the Sega logo before the title screen if the game detects it is being played on a Japanese system.
Configuration
On the title screen, press 
on the second Control Pad.
